研究内容

ご自慢の頬ひげであたりを探索し,何かをみつけては齧りつくネズミたち。動物にとって感覚は,生きるために必要な能力,というより生き甲斐そのものだ。頬ひげの動きや歯のかみ合わせを検出する機械受容器には,さまざまな形をしたものが知られ,その発見者として,顕微鏡学の大先輩たちが名を連ねている。こうした結合組織の感覚装置にきまって現れるのが,終末シュワン細胞である。終末シュワン細胞は,丸い細胞体(Fig.1のS)の一端から長い突起をのばし,その先が分かれて一次感覚ニューロンの軸索終末を被うグリア薄板をつくる(Fig. 1)。

じつは,各種機械受容器の形を特徴づけるのは軸索終末ではなく,グリア薄板の縁から周囲結合組織にのび出た小さな副突起(Fig.1の矢印)なのだ。歯のかみ合わせの感覚装置ルフィニ終末で,終末シュワン細胞の薄板突起(青)と2枚の薄板にサンドイッチされた軸索終末(黄)の関係を調べてみよう(Fig. 2)。

Fig. 2左は走査電顕,右は透過電顕。グリア薄板から舌状の副突起がでて,歯根膜の膠原線維をしっかりとつかんでいる。ラット頬ひげの動き受容器はどうだろう(Fig.3)

終末シュワン細胞(青)の副突起は粘液状物質の中を紐のようにのび,毛包基底膜の裏面や膠原線維上に終足を下す。機械受容器の軸索終末は終末シュワン細胞の副突起を介し,周囲結合組織の外力による変形を感受しているのかもしれない(Fig. 4)。

しかし,ニューロンでもないシュワン細胞が,どのようにして,機械刺激の影響を受けやすい場所に一定の形の副突起をつくるのだろう。私たちは今,終末シュワン細胞の刺激入力依存的な形態変化と,細胞自身の刺激応答性との関連を調べている(Fig. 5)。

ラットの頬ひげ毛包から分離した槍型終末にCa2+指示薬Fluo-4を負荷して共焦点顕微鏡のステージにセットし,小さなガラス針で軽い接触刺激を与えたときの細胞応答をFluo-4蛍光強度変化としてタイムラプス記録したのがFig. 5である。蛍光強度増加分を赤色で示す。刺激後,槍型終末(L)を被う薄板突起に限局した一過性の細胞内Ca2+濃度上昇がおこる。Sは終末シュワン細胞体。皮膚感覚装置のさまざまな姿の意味を探り,奮闘中である。
